摘要:
为了制备鹅星状病毒(goose astrovirus,GAstV)刺突(Spike)蛋白的单克隆抗体,并对获得的单克隆抗体的生物学特性进行鉴定,将大肠杆菌表达的截短Spike蛋白免疫BALB/c小鼠,经细胞融合和单克隆抗体的筛选来获得阳性杂交瘤细胞,并进一步利用免疫印迹和间接免疫荧光技术进行单抗的鉴定;采用ELISA技术进行单克隆抗体亚型、抗体效价的测定;同时根据GAstV XX株 Spike蛋白序列合成重叠的多肽库进行单抗识别的抗原表位鉴定。共获得3株阳性杂交瘤细胞,分别命名为1A5、2B4、12E6;免疫印迹和间接免疫荧光实验显示3株单克隆抗体均与Spike蛋白具有良好的免疫反应,其中2B4、12E6可与GAstV发生反应;亚型鉴定结果显示单抗12E6、2B4为IgG2b亚类,单抗1A5为 IgG2a亚类;轻链均为κ链;敏感性结果显示,抗体效价都在1 ∶[KG-*3]51 200以上;抗原表位的鉴定结果表明单抗1A5识别的抗原表位为(EP16)ELRNRLNIADGDYVI,单抗2B4识别的抗原表位为(EP21)AGDSNPGETFQNFKM。本研究成功制备了针对GAstV Spike蛋白的单克隆抗体,并鉴定出2个新的抗原表位,为建立快速、简便的GAstV检测方法奠定了基础。
