摘要:
为探究甜瓜果实在采后贮藏过程中脂氧合酶对果实后熟软化生理及相关基因表达的影响,以西州蜜17号为试验材料,通过对甜瓜果实贮藏期间脂氧合酶活性、乙烯释放量、果实硬度、呼吸强度、果胶酶活性、淀粉酶活性以及相关基因表达量进行测定,研究甜瓜果实采后脂氧合酶代谢、乙烯产生、后熟软化生理及相关基因表达的作用关系。分别将甜瓜果实在20 ℃贮藏(对照组)、经1-MCP处理后20 ℃贮藏、2 ℃贮藏,分析甜瓜果实中LOX活性与乙烯释放量的变化趋势。结果表明,在甜瓜果实后熟软化过程中,LOX活性上升先于乙烯释放量的增加,而在乙烯释放量与呼吸强度跃变后,LOX活性逐渐下降。同时,果实中果胶酶与淀粉酶活性及其基因表达量在跃变现象发生后显著增加,果实硬度随之迅速下降,说明果胶酶与淀粉酶基因表达增强和酶活性升高是甜瓜果实发生软化的关键因素。此外,LOX活性升高能够启动乙烯释放量的增加,促使果实发生呼吸跃变并进入后熟阶段,但乙烯对LOX却没有调控作用。由此可知,LOX活性对果胶和淀粉的变化没有直接影响,而是通过启动乙烯的释放来调节果胶和淀粉的代谢反应,使果实发生软化。
