摘要:
采用硫酸铵沉淀及Sephadex G-100凝胶过滤柱、DEAE-Sepharose阴离子交换柱、SP SephadexC-25阳离子交换柱层析等步骤,对豆芽中磷酸吡哆醛(PLP)水解酶进行分离纯化。结果表明:黄豆芽PLP水解酶被纯化了220.19倍,得率为23.65%。最适温度为50 ℃,最适反应pH值为5.5;Mg2+、Zn2+、Mn2+ 等对该酶有激活作用,Cu2+、Hg2+及金属离子螯合剂EDTA对该酶有抑制作用,而Ca2+对该酶活性无明显影响。在最适反应条件下,测得反应底物磷酸吡哆醛和磷酸吡哆胺(PMP)的Km分别为0.27、0.41 mmol/L。
