摘要:
为了研究蛋白酶体抑制剂MG132对小鼠卵母细胞排卵后老化的调控作用,找到抑制卵母细胞老化的方案,通过孤雌激活和胚胎发育试验系统研究了MG132在不同温度下对小鼠卵母细胞老化的调控作用,并通过体外受精发育以及细胞免疫组化试验,分别研究了15 ℃时MG132对卵母细胞受精后胚胎发育能力、皮质颗粒分布与BCL2蛋白水平的影响。结果表明,15 ℃时卵母细胞在添加1 μmol/L MG132的HCZB中培养12 h,然后在37 ℃恢复6 h,是筛选的抑制卵母细胞老化的优化方案,说明一定低温联合适当浓度的蛋白酶体抑制
