摘要:
原钙黏蛋白(protocadherin)是广泛分布于中枢神经系统的一类跨膜蛋白,是钙黏蛋白家族中最大的亚族。成簇Pcdh基因包含3个串联的基因簇——Pcdh α、β、γ,其中Pcdh αγ分别由14个和22个可变区及各自共同的恒定区组成。这种特殊蛋白结构决定了原钙黏蛋白各亚型在功能上既相似又不完全相同,为了系统研究原钙黏蛋白各个亚型的功能,本研究对原钙黏蛋白家族氨基酸序列保守性进行了系统性分析,并据此设计特异性的引物来完成原钙黏蛋白各个亚型的克隆;并以原钙黏蛋白α1基因为例,通过PCR方法成功将其从小鼠大脑中克隆并构建到带有Myc标签的真核表达载体pcDNA3.1上;利用脂质体转染方法将该基因成功表达于293T细胞。本研究结果将为后续其他亚型的克隆建立技术平台。
