摘要:
应用DNA混池测序法检测藏羊HIF-1α基因CDS区的多态性,并采用生物信息学方法对该基因及其编码蛋白的基本理化性质、疏水性、信号肽、二级结构等方面进行预测和分析。结果表明,在藏羊群体中发现HIF-1α基因CDS区有4个SNP,其中901G→A为错义突变,其余为同义突变;HIF-1α基因包含1个2 295 bp的开放阅读框,编码764个氨基酸,其编码蛋白属于亲水性蛋白;二级结构主要以无规则卷曲和α基因螺旋为主;HIF-1α基因编码产物氨基酸同源分析表明,藏羊HIF-1α编码的氨基酸与山羊、牦牛的遗传距离较近,具有高度同源性。
