摘要:
为研究青钱柳多糖对高脂血症小鼠抗脂质过氧化相关基因表达的影响,将建模成功的高脂血症小鼠随机分为青钱柳多糖高、中、低剂量组,辛伐他汀组,高脂模型组和空白对照组,其中前4组分别灌胃400、200、100 mg/kg青钱柳多糖水溶液,4 mg/kg辛伐他汀水溶液,另2组灌胃等量无菌水,每组16只,连续喂养4周。试验结束后采用逆转录PCR(RT-PCR)测定肝脏、脂肪组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)基因的mRNA表达水平,探讨青钱柳多糖抗脂质过氧化机制。结果表明,不同剂量青钱柳多糖可使SOD、GSH-Px、CAT mRNA表达量显著或极显著上升(P<0.05或P<0.01),其中肝脏组织中SOD、CAT表达量最大增幅分别达1309%、108.6%,GSH-Px表达量未出现显著变化;脂肪组织中SOD、GSH-Px的最大上调幅度分别为507%、1000%,而CAT表达量无显著差异。结果显示,青钱柳多糖可调节高脂血症小鼠抗氧化相关基因的表达,增强其机体抗脂质过氧化能力。
