摘要:
Ldhc基因仅在哺乳动物成熟的睾丸和精子中表达,并在哺乳动物的睾丸中普遍存在剪接体现象,但是Ldhc基因在肌肉和心脏组织中的表达情况以及与睾丸中该基因的表达丰度比较尚不清楚。因此,以小鼠为研究对象,首先检测小鼠肌肉和心脏组织中Ldhc基因的表达类型,然后采用荧光定量和Western Blot比较分析3个组织中Ldhc基因和蛋白的表达丰度。结果表明,在小鼠肌肉和心脏组织中未检测到cDNA全长,却发现2种Ldhc剪接体,参照小鼠Ldhc基因结构分析了选择性剪接体的结构;剪接体1包含完整酶的NAD+结合结构域和酶的活性中心,有可能表达蛋白;剪接体2缺失第4、7外显子。对小鼠肌肉、心脏和睾丸组织乳酸脱氢酶(LDH)同工酶的电泳分析未检测到潜在的Ldhc剪接体的表达。荧光定量检测发现,Ldhc基因在睾丸组织中的表达量要显著高于肌肉和心脏组织,而Western Blot仅在睾丸组织中检测到LDHC4蛋白的表达。以上结果表明,小鼠Ldhc基因的选择性剪接机制也存在于肌肉和心脏组织中,推测可能是调控该基因在肌肉和心脏组织中沉寂的方式之一。
