摘要:
以云烟87为试验材料,采用烘烤比较试验,设置3个处理,即CK,云南当地常规烘烤工艺;T1,变温幅度±1.0 ℃、变温频率3 h;T2,变温幅度±1.5 ℃、变温频率5 h,探索密集烤房变温调控对烤烟上部叶氮代谢生理的影响。利用曲线估算、通径分析对烟叶可溶性蛋白质、游离氨基酸含量及氮代谢相关酶活性进行系统分析,研究烘烤过程中变黄期变温处理对烟叶蛋白质降解机理的影响。结果表明,2个变温工艺处理下,烟叶中性蛋白酶、内肽酶活性呈“M”形曲线变化,转氨酶活性呈先升后降再升的变化趋势,与常规工艺处理有所差异;烘烤变黄结束时,T1变温处理烟叶的中性蛋白酶、内肽酶、谷草转氨酶、谷丙转氨酶活性分别为0.058 6、0.081 6、31.113 9、36.714 8 U/g,高于或显著高于CK、T2处理。在变温幅度1 ℃和变温频率3 h处理条件(T1)下,对烟叶可溶性蛋白质、游离氨基酸含量影响的主要因素是谷草转氨酶、内肽酶活性;烘烤变黄结束时,T1变温处理下的可溶性蛋白质含量最低,游离氨基酸含量为380.667 9 μmol/g,显著高于CK、T2处理。对密集烤房烘烤变黄期变温幅度和变温频率进行合理调控,有利于氮代谢相关酶活性维持较高水平,促进蛋白质的充分降解和游离氨基酸的有效积累,可明显影响烟叶品质。
