摘要:
细菌染色体上的mnt/hepn操纵子构成毒素-抗毒素系统(TA,toxin-antitoxin system)新家族,但该家族成员的生化及遗传特征有待阐明。为了证明集胞藻PCC6803染色体上基因ssl2749/sll1411的转录调控作用,以无启动子的β-半乳糖苷酶基因(lacZ)为报告基因,构建了lacZ转录融合重组质粒,通过测定含重组质粒的大肠杆菌细胞的β-半乳糖苷酶活性,确定ssl2749/sll1411编码产物对其启动子活性的调控作用。结果表明,蛋白Ssl2749使β-半乳糖苷酶活性显著增加,提示Ssl2749可激活PTA启动子转录活性,蛋白Sll1411抑制Ssl2749的转录激活作用。因此,Ssl2749可能与PTA中的调控序列结合调控ssl2749/sll1411操纵子的转录,Sll1411对Ssl2749转录激活活性的抑制作用提示二者之间存在相互作用。
