摘要:
海棉7124号棉花种子在常规花盆土壤中种植发芽,待棉花幼苗长至2叶1心时,将其随机分为对照组和试验组,分别置于常温(25 ℃)和低温(5 ℃)植物培养箱中,光照/黑暗12 h交替培养,每天适量浇水。各组分别在0、12、24、48、96 h取根尖组织,并提取 RNA,以逆转录、荧光定量PCR法测定亚型乙醇脱氢酶(ADHa)和甜菜碱醛脱氢酶(BADH)基因的表达。结果显示,在各时间点下,低温组ADHaBADH表达量均明显高于常温组。说明幼苗期棉花根系ADHaBADH表达量在低温胁迫条件下升高与低温抗逆性有关。
