摘要:
对紫薇属植物细胞核核糖体DNA转录间隔区(nrDNA ITS)序列进行分析,为紫薇植物的鉴定提供分子依据。通过提取紫薇幼苗总DNA,对nrDNA ITS序列进行PCR扩增测序,应用软件BioEdit、DNAMAN进行序列分析,MEGA计算遗传距离,构建基于K2P模型的NJ系统发育树。测序得到2条nrDNA ITS序列,长度均为615 bp。序列分析结果显示,序列1与紫薇ITS序列相似性达99.03%,遗传距离为0.003,聚类分析归为一支;序列2与毛紫薇ITS序列相似性达98.55%,遗传距离0.007,聚类分析归为一支。试验结果说明,基于nrDNA ITS序列分析法,能够对紫薇属植物进行准确的分子鉴定,从而为紫薇属的种类鉴定和种间亲缘关系提供分子生物学理论依据。
