采用福美双诱导建立肉鸡胫骨软骨发育不良(tibial dyschondroplasia,TD)模型,研究甲状旁腺素相关肽-印第安刺猬蛋白(PTHrP-Ihh)信号轴在肉鸡发生胫骨软骨发育不良中的调控机制。100羽1日龄健康Ross308肉鸡经1周预饲后,随机分为2组,对照组饲以基础日粮,试验组在饲喂8、9 d时饲以基础日粮并添加100 mg/kg福美双,之后继续饲以基础日粮;于15日龄时采集组织样本,进行检测。结果显示,与对照组比较,试验组体质量极显著降低,但TD 发生率极显著升高;胫骨与肱骨的骨长度、骨密度,胫骨与股骨的骨强度均显著或极显著降低,而肱骨的骨指数显著增强;TD肉鸡胫骨生长板软骨组织Ihh、PTHrP的mRNA表达极显著下降,ColⅩmRNA表达极显著上升。试验表明,PTHrP-Ihh信号轴调控紊乱,会影响生长板软骨细胞的分化和ColX表达,导致肉鸡形成胫骨软骨发育不良。
