摘要:
在紫外诱变的基础上,采用单因素试验和正交试验对侧孢短芽孢杆菌(Brevibacillus laterosporus) M64发酵生产几丁质酶的培养条件和培养基进行优化,并对粗酶液的酶学特性进行了初步研究。结果表明,碳氮源的种类与浓度、培养温度、pH值以及表面活性剂吐温80浓度对该菌株产几丁质酶有显著影响。突变株使用优化后的培养条件,其发酵上清液中几丁质酶活力达到55.19 U/mL,为未优化前的2.56倍;粗酶液最适催化温度为60 ℃,且在80 ℃预处理2 h后仍具有降解几丁质的能力,表现出了良好的热稳定性。
