摘要:
病毒同源重组是以同源臂为基础的将目的基因整合到病毒基因组中的技术。将表达盒串联到转移载体,通过同源重组构建重组病毒从而实现病毒基因改造、外源基因表达以及重组活载体疫苗制备等。就重组病毒的优缺点、构建原理及应用进展进行了概述,对重组病毒构建的难点进行了分析,全面系统地阐述了重组病毒的构建模式,针对基因框(启动子-报告基因-目的基因-调控元件-终止子)给出具体的构建方案,同时对其未来发展前景进行了展望,以期为今后重组病毒的构建提供理论基础。
