摘要:
对具有典型病症的染病草鱼进行了病原菌分离鉴定。从其肝脏中分离得到病原菌菌株FK-1,经VITEK-32全自动细菌鉴定仪鉴定为荧光假单胞菌。同时,研究了荧光假单胞菌FK-1侵染草鱼对草鱼机体免疫因子的影响,通过腹腔注射荧光假单胞菌FK-1,在侵染后不同时间点取草鱼的血液和肝脏组织,分别采用分光光度计法和ICP-AES法检测血清和肝脏中的铁元素含量,同时采用实时荧光定量PCR法对铁调素基因(hepc)、白细胞介素基因(il-6)、JAK/STAT信号通路的蛋白质酪氨酸激酶3基因(jak3)、信号转导和转录激活因子3基因(stat3)等基因表达量进行检测。结果表明,荧光假单胞菌FK-1侵染草鱼后,草鱼机体血清中铁元素浓度明显降低,在24、48 h时显著低于对照组,总铁结合力有所上升,但没有达到显著水平;肝脏中铁元素含量相对于对照组没有达到显著性差异水平,但仍有明显升高。肝脏中hepc基因的表达量显著上升;il-6、jak3和stat3基因表达量开始均明显上调,随后上升不明显,但各时间点的表达量均高于对照。表明草鱼机体通过调节铁调素基因(hepc)等铁元素相关基因的表达,进而降低自身游离铁元素和增加储存铁元素来应对荧光假单胞菌FK-1的侵染。
