摘要:
灵芝多糖(GLP)是灵芝中主要功能性活性成分,具有增强免疫力、抗氧化、抗炎、抗肿瘤、降血糖和降血脂等功效。采用大肠杆菌内毒素脂多糖(LPS)诱导小鼠乳腺炎,研究灵芝多糖对小鼠乳腺炎的保护作用。试验选取刚分娩的健康雌性小鼠75只,分为空白对照组、LPS组、LPS+2% GLP组、LPS+4% GLP组和LPS+8% GLP组,LPS+2% GLP组、LPS+4% GLP组、LPS+8%GLP组分别灌胃2%、4%、8%的灵芝多糖溶液,空白对照组和LPS组灌胃等量的生理盐水。注射LPS后,测量小鼠体温变化,24 h后取乳腺制作石蜡组织切片,并测定乳腺中的IL-1β、IL-6、TNF-α含量和MPO活性。结果表明,与空白对照组相比,LPS组小鼠体温变化显著,乳腺组织切片出现明显的中性细胞浸润现象,IL-1β、IL-6、TNF-α含量和MPO活性显著上升;LPS+2% GLP组、LPS+4% GLP组、LPS+8%GLP组与LPS组相比,小鼠体温变化有显著改善,18 h后LPS+4% GLP组和LPS+8% GLP组与LPS组差异显著(P<0.05);与LPS组相比,LPS+2% GLP组、LPS+4% GLP组、LPS+8%GLP组小鼠乳腺中性细胞浸润现象明显减轻,LPS+4% GLP组和LPS+8% GLP组的TNF-α、IL-1β含量显著低于LPS组(P<0.05);LPS+4% GLP组和LPS+8% GLP组中IL-6含量显著低于LPS组(P<0.05),LPS+2% GLP组与LPS组差异不显著(P>0.05)。上述结果表明,添加4%、8%灵芝多糖可以减轻由LPS引起的小鼠乳腺炎反应。
