摘要:
以1株筛选自盐浓度为21%海盐水的中度嗜盐菌Salinivibrio sp. MK070915为研究对象,该菌株可产胞外蛋白酶。以Gibbons培养基为基底,采用单因素试验和最陡爬坡试验相结合的方法对影响嗜盐菌产蛋白酶能力的重要培养基成分和工艺条件进行筛选,确定主要影响因子及其取值范围;并在此基础上,通过Design-Expert软件对主要影响因子进行四因素三水平响应面试验设计,以蛋白酶活性为响应值优化菌株产酶的培养条件,并验证响应面模型预测值与实测值的一致性。结果表明,最优培养条件如下:以Gibbons培养基为基底,NaCl浓度为5%,pH值为8,培养温度为20 ℃,碳源为麦芽糖,含量为1.85%,蛋白胨的含量为1.17%,KCl的含量为1.0‰,柠檬酸钠的含量为5‰,经响应面分析得到的蛋白酶活性为44.09 U,与预期值44.22 U相比,差异为0.299%。结果丰富了嗜盐蛋白酶的研究和开发依据,为高盐环境下食品工业催化和农业制剂催化提供了理论基础。
