摘要:
高含量的维生素C是刺梨果实的重要性状,挖掘与维生素C含量相关联的基因序列变异及单倍型对刺梨的品种改良具有指导意义。在21份野生刺梨资源中对GGPDHAR基因进行PCR扩增和测序,分析基因序列多态性及其与果实维生素C含量的相关性。GGP基因在21份野生刺梨中共扩增出长度为2 035 bp的同源序列,其中包含22处变异位点,产生15种单倍型,在0.01显著性水平上,3个单核苷酸多态性(SNP)及Hap15与刺梨果实高含量维生素C相关;在0.05显著性水平上,3个SNP及Hap7与刺梨果实低含量维生素C相关;GGP基因单倍型存在网状进化。DHAR基因在21份野生刺梨中共扩增出长度为1 609 bp的同源序列,其中包含69处变异位点,产生9种单倍型,在0.05显著性水平上,8个SNP、9个InDel及Hap9与刺梨果实中低含量维生素C相关,1个SNP及单倍型Hap4~Hap6与刺梨果实中高含量维生素C相关;DHAR基因单倍型呈现扇形进化。GGP基因具有更高的单倍型多样性,DHAR基因具有更高的DNA多态性;GGP基因特异单倍型Hap15DHAR基因特异单倍型Hap4共同存在于刺梨中时,可以使果实维生素C含量表现为高的状态;而GGP基因特异单倍型Hap7DHAR基因单倍型Hap9同时存在于刺梨中时,可以使果实维生素C含量表现为低的状态。
