摘要:
将黑曲霉属XynB基因和猪RELMβ基因核心启动子区克隆到pcDNA3.1(-)中,构建携带绿色荧光和Myc双标记的肠道特异表达载体pcDNA3.1-RELMβ-XynB-Myc-GFP。利用脂质体介导将载体转染人结肠癌细胞(HT29)和人肝癌细胞(Bel7402),荧光显微镜检测发现,该载体可在HT29细胞特异表达绿色荧光蛋白;对转染该表达载体的HT29细胞进行RT-PCR检测和WB分析,结果表明,XynB基因在HT29细胞中正常转录,并且在细胞内检测到目的蛋白表达。
