摘要:
为研究湖北地区禽腺病毒4型遗传进化情况及其Fiber2蛋白在乳酸菌中的表达情况,将采集于湖北省某养鸡场的病变组织样处理后接种至LMH细胞,分离出2株禽腺病毒,通过PCR鉴定和Hexon基因测序分析表明为禽腺病毒4型,并命名为HBJZ2021A和HBJZ2021B,遗传进化树分析结果表明,其与国内山东、浙江和北京等地区的毒株亲缘关系较近,处于同一进化分支。以其基因组DNA为模板扩增Fiber2基因并克隆至乳酸菌表达载体pNZ8148,通过酶切和测序表明成功构建了重组表达载体pNZ8148-Fiber2,并通过电转转化至NZ 9000乳酸菌,采用不同浓度梯度的Nisin进行诱导表达,SDS-PAGE及Western-Blot分析结果显示,成功在上清中表达了Fiber2蛋白。该研究结果为湖北地区禽腺病毒4型的流行情况及其口服疫苗的开发提供了参考依据。
