基于ITS序列对紫薇的分子鉴定-《江苏农业科学》

摘要:: 对紫薇属植物细胞核核糖体DNA转录间隔区(nrDNA ITS)序列进行分析，为紫薇植物的鉴定提供分子依据。通过提取紫薇幼苗总DNA，对nrDNA ITS序列进行PCR扩增测序，应用软件BioEdit、DNAMAN进行序列分析，MEGA计算遗传距离，构建基于K2P模型的NJ系统发育树。测序得到2条nrDNA ITS序列，长度均为615 bp。序列分析结果显示，序列1与紫薇ITS序列相似性达99.03%，遗传距离为0.003，聚类分析归为一支；序列2与毛紫薇ITS序列相似性达98.55%，遗传距离0.007，聚类分析归为一支。试验结果说明，基于nrDNA ITS序列分析法，能够对紫薇属植物进行准确的分子鉴定，从而为紫薇属的种类鉴定和种间亲缘关系提供分子生物学理论依据。
