摘要:
以血清1型鸭甲型肝炎病毒(DHAV-1) SH株3C基因为序列设计1对特异性引物,RT-PCR方法进行扩增,克隆入原核表达载体pET-32a,转化大肠杆菌BL21(DE3),在IPTG诱导下重组蛋白并获得成功表达,该重组蛋白分子量约为38 ku,能与多聚组氨酸标签单抗发生特异性反应。将目的蛋白切胶免疫ICR小鼠,制备重组蛋白的多抗血清,经Western-blot分析证明,制备的抗血清能够与DHAV-1感染的SPF鸡胚尿囊液总蛋白发生特异反应。3C基因在大肠杆菌中可获得成功表达,制备的多抗血清可用于3C蛋白的检测。
