摘要:
以桃叶卫矛(Euonymus bungeanus Maxim.)为材料,选用茎尖作为外植体进行超低温保存,以期探索桃叶卫矛种质资源保存的新途径。桃叶卫矛茎尖超低温保存中茎尖最适长度为2~3 mm;在预培养阶段,预培养液中蔗糖最适浓度为0.4 mol/L,最佳预处理时间为2 d;在装载液处理阶段,最佳装载时间为10 min;在PVS2溶液处理阶段,最佳处理时间为60 min;在卸载液处理阶段,最佳处理时间为10 min;当茎尖洗涤完毕后应将其接种于1.0 mg/L 6-BA的MS培养基上进行恢复培养,暗培养7 d后转入正常光照下进行培养,茎尖成活率可达70%以上。
