摘要:
先采用二乙氨基乙基-纤维素(diethlaminoethyl cellulose,简称DEAE-纤维素)柱对党参多糖进行分离纯化;然后,分别采用高效离子色谱和高效排阻色谱分析分级产物的单糖组成和分子量;最后,采用DPPH自由基、超氧负离子的清除能力试验以及D-半乳糖(D-Gal)诱导的小鼠衰老模型试验分析党参多糖的抗衰老活性。结果发现,党参多糖经DEAE-纤维素柱分级后得到CPNP-1和CPNP-2 2个级分,均主要由鼠李糖、半乳糖醛酸、半乳糖和阿拉伯糖构成,分子量分别约为51、23 ku。体外抗氧化和体内抗衰老活性研究结果表明,CPNP-2具有较强清除DPPH自由基和超氧负离子的活性,半最大效应浓度EC50分别为1.5、0.9 mg/mL,并且能够有效缓解衰老模型小鼠血清的超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性下降情况。结果表明,本试验建立的分级方法适用于党参多糖中抗衰老活性成分的分离纯化,促进党参多糖类抗衰老类药品的开发利用。
