摘要:
以桃为材料,研究了影响cDNA-AFLP反应体系的几个关键因素, 建立了适宜桃的cDNA-AFLP分析体系, 得到了较为清晰可辨的cDNA-AFLP指纹图谱。结果表明,改良CTAB法提取的RNA 较完整,纯度较高;反转录试剂盒形成的双链cDNA经EcoRⅠ和MseⅠ完全酶切后,16 ℃连接过夜;20 μL的体系中,连接产物2.0 μL作为预扩增的模板,循环数为30,预扩增产物稀释10倍作为选择性扩增的模板,扩增结果较好。
