摘要:
采用RT-PCR和TA克隆技术,从水稻品种日本晴中扩增出类整合素候选基因(integrin-like protein,ILP)OsILP的cDNA片段。经测序鉴定,克隆获得的目的片段长为1 167 bp,包含完整的CDS,编码388个氨基酸,预测分子量43 ku。进而将该片段连接至表达载体pTCK303中,通过PCR鉴定与酶切验证,结果表明成功构建了pTCK303-OsILP过表达载体。并将表达载体质粒转化为农杆菌EHA105,再通过农杆菌介导将其导入水稻日本晴的愈伤中,经遗传转化,阳性鉴定,获得了过表达转OsILP基因水稻株系。
