摘要:
根据已知的其他物种白藜芦醇合酶cDNA保守序列设计引物,用RT-PCR技术从桑葚中扩增获得白藜芦醇合酶基因部分cDNA序列,再用RACE技术获得其两端序列,并拼接得到完整的1 442 bp白藜芦醇合酶基因。经序列分析发现,桑树白藜芦醇合酶基因开放阅读框长1 170 bp,编码389个氨基酸,氨基酸序列含有芪合酶家族特征信号区GVLFGFGPGLT和活性中心序列GCFAGGTVLR;该基因与花生芪合酶基因的核苷酸序列同源性高达82.82%,氨基酸序列的同源性高达87.15%。
