摘要:
对兴国红鲤精液的超低温冷冻保存技术进行了研究。以解冻后精子的寿命和运动力为参数,分别探讨了不同组合的稀释液、抗冻剂以及冷冻程序对兴国红鲤精液进行超低温冷冻保存的保护效果。试验结果表明,选用Ringer液为稀释液,8% DMSO为抗冻剂,按照程序-80 ℃平衡15 min,后保存于-196 ℃液氮中,37 ℃水浴100 s解冻,精液解冻激活后镜检,精子活力达到(83±9.6)%,寿命在(86.25±24.7) s,接近鲜精的水平。本研究结果为保护淡水鱼类的种质资源提供了理论基础。
