摘要:
为提高蛋白质的稳定性,便于运输和延长储存时间,冷冻干燥是目前公认最可行的技术之一。通过对猪重组抑制素蛋白冷冻干燥后的外观、色泽、复水性以及SDS-PAGE电泳检测,对27种常用冻干保护性物质进行筛选。结果表明,可溶淀粉和β-环糊精的保护效果较好,表现在外形饱满无坍缩、色泽细腻白亮、复溶后无沉淀、电泳检测发现蛋白条带无降解。将筛选出的2个保护剂制备成冻干样品放置于37 ℃烘箱中,进行加速稳定性试验。结果表明,在37 ℃放置4个月后,冻干蛋白样品保持形态色泽不变,复溶后无沉淀,蛋白无降解现象。
