摘要:
为了对小麦成熟胚的再生优化体系进行培养效果评价及筛选高再生率基因型,以7个小麦品种成熟胚为材料,采用常规和优化2种培养基体系,研究3种胚处理方式对成熟胚出愈率、分化率及再生率的影响。结果表明:优化培养基体系组织培养效果显著优于常规培养基体系;胚处理方式与基因型之间可能存在一定的互作关系,特定基因型品种采用不同胚处理方法,其组织培养效果差异明显,且不同品种的最适胚处理方式可能不同。在供试的7个品种中,矮抗58、济麦22、百农419、百农160和周麦22在经完整胚法处理,分化率均达到50%以上,再生率均达到25%以上,其中以矮抗58和百农160最优,再生率分别达到51.74%、43.84%。而采用纵半切略带胚乳法时,宝丰7228和中麦175的分化率和再生率均较完整胚法有了较大提升,分别达到62.89%、56.79%和26.88%、36.75%。说明特定基因型品种须要找到最适合的胚处理方法,以充分挖掘其再生能力,获得的高再生率小麦品种可用于遗传转化研究。
