摘要:
通过构建含平菇内源gpd启动子、红豆杉中TwDBAT基因的真核表达载体pgFvs-TwDBAT,建立高效的平菇原生质体制备与再生体系;采用PEG介导法,将真核表达载体pgFvs-TwDBAT、pgFvs-hph共转化高温型平菇菌株F7E原生质体,获得可以异源表达TwDBAT基因的平菇菌株。PCR和Southern杂交鉴定结果表明,TwDBAT基因整合进平菇基因组中;经继代培养,TwDBAT基因在转化子中可稳定遗传。RT-PCR结果表明,转化子在mRNA水平上转录了TwDBAT基因。HPLC结果显示,TwDBAT酶可以催化底物10-去乙酰巴卡亭Ⅲ(10-DAB)。
