摘要:
利用RACE技术首次从荸荠中克隆得到PPO基因全长cDNA序列,并分析其在鲜切荸荠贮藏过程中的表达变化。克隆得到的荸荠PPO基因cDNA序列全长为2 006 bp,开放阅读框长度为1 734 bp,编码577个氨基酸,命名为CwPPO(登录号:MG702489)。预测分析表明,CwPPO编码蛋白质的分子量为64.86 ku,分子式为C2891H4417N795O873S18,理论等电点为6.50,属亲水性蛋白。CwPPO蛋白可能定位于线粒体基质空间,具有3个典型的保守结构域和2个铜离子结合域。此外,预测发现CwPPO有20个丝氨酸磷酸化位点、7个苏氨酸磷酸化位点及6个酪氨酸磷酸化位点,其二级结构以不规则卷曲为主。系统发育分析显示,CwPPO蛋白与菠萝、油棕有较近的亲缘关系。荧光定量分析发现,CwPPO在鲜切荸荠中的初始表达水平较低,但在贮藏后期表达量快速提高。
