摘要:
采用硫酸铵盐析、Sephadex G-75柱层析等方法得到山药多酚氧化酶(PPO),纯化倍数为52.5倍,活力回收率为16.45%。结果表明:PPO最适温度为35 ℃,最适pH值为5.0。温度55 ℃时PPO较为稳定,酶活性为55%;pH值6.5~7范围内酶活性变化不大,约为80%。山药PPO催化合成茶黄素,产量为28.1%。茶黄素-3-没食子酸酯(TF-3-G)、茶黄素-3′-没食子酸酯(TF-3′-G)、茶黄素双没食子酸酯(TFDG)、茶黄素(TF)含量分别为12.4%、2.78%、9.46%、0.85%。
