摘要:
为筛选淀粉酶高产菌株,通过比较水解圈与菌落的直径比及3,5-二硝基水杨酸(DNS)显色法所测酶活性高低,筛选得到高产淀粉酶能力的菌株WB-4。通过单因素试验,得到该菌株的最优碳源为可溶性淀粉,最优氮源为蛋白胨。利用Plackett-Burman进行显著因素筛选,发现可溶性淀粉含量、蛋白胨含量、温度为显著因素。利用响应面法对显著因素进行优化,得到菌株的最优培养条件为可溶性淀粉含量18.81 g/L,蛋白胨含量10.02 g/L,温度 31.12 ℃,淀粉酶活性 365.12 U/mL,酶活性提高了1.33倍。
