摘要:
为从分子水平研究猪脂肪合成过程,对脂肪合成关键酶乙酰辅酶A羧化酶-α基因(ACACA)进行分析。采用生物信息学的方法,查找猪ACACA基因的ORF框,对基因CDS序列的限制性酶切位点进行分析,并对其编码蛋白质的理化性质、二级结构、三级结构、拓扑结构、结构功能域进行预测分析。结果表明,猪ACACA基因CDS区全长 7 041 bp,其编码蛋白质含2 346个氨基酸。二级结构以α螺旋与无规则卷曲为主,局部出现β转角,三级结构显示结果与二级结构预测结果相符。该蛋白质为亲水性蛋白质,N端无信号肽,为非分泌蛋白质;不含跨膜结构区,未定位于膜内,预测其为膜外区蛋白质;亚细胞定位结果显示该蛋白质为非细胞器蛋白质,是胞质蛋白质的可能性最大;功能域分析发现该蛋白质序列含有BC结构域、CT结构域、CPSase 大亚基N端结构域、ATP-grasp折叠结构域、生物素基/硫辛酰基结合域等及一些蛋白质基序。该结果将为进一步研究猪ACACA基因与脂肪合成的关系提供基础资料。
