摘要:
以穿心莲提取物为试验原料,采用动态轴向压缩制备色谱技术进行分离纯化,以制备高纯度穿心莲内酯。制备柱装填纯化材料:硅胶基质C18 10 μm,装填质量300 g,实测柱床规格50 mm×250 mm。以甲醇水为流动相进行纯化工艺优化,结果表明,甲醇水体系下水相添加0.05 mol/L乙酸铵能促进穿心莲内酯的溶解,该体系下穿心莲内酯最佳纯化工艺条件:流动相55%甲醇水+45% 0.05 mol/L乙酸铵缓冲液,流速60.0 mL/min;检测波长254 nm,温度为室温。
